Ultimately, the lack of targeted therapy options served as the basis for the CodeBreak 100 study, says Price. To date, no 

-specific targeted therapies have been approved for patients with GI cancers, Price explains. However, options are available for patients with 

-mutant GI cancers like colorectal cancer and non–small cell lung cancer.

Patients enrolled to the ​ongoing phase 1 CodeBreaK 100 trial ​were heavily pretreated, Price says. As such, patients had to undergo careful screening to ensure that they were fit to tolerate another course of treatment. This was one of the primary challenges faced during the phase 1 portion of t​he study, Price says.

In addition, ​eligible patients had to have 

 is a rare mutation in the GI cancers space and only about 2% to 3% of patients with colorectal cancer (CRC) harbor 

 alterations. In the 50% of patients with CRC that express 

 mutations, a commonly mutated gene across multiple tumor types, only about 5% to 6% will ​harbor

 alterations. As such, screening to identify these rare mutations has been an additional challenge ​in the CodeBreaK 100 trial, Price concludes.

Timothy Price, MBBS, DHthSc, FRACP, senior consultant medical oncologist and director of Medical Oncology and Clinical Cancer Research at The Queen Elizabeth Hospital in Adelaide, discusses the safety and efficacy of AMG 510 in patients with advanced gastrointestinal cancers.

The phase 1 results of the CodeBreaK 100 trial were presented during the 

 and focused on the benefit of AMG 510 in patients with colorectal cancer (CRC) and other gastrointestinal cancers. Results showed that the drug was safe, with few toxicities associated with single-agent AMG 510; this was an important finding for a phase 1 study, Price explains. For the patients with colorectal cancer specifically, a signal of activity was observed. The drug elicited a promising response rate and the majority of patients achieved stable disease, adds Price.

Notably, the patients enrolled on the study were heavily pretreated; however, they were able to achieve impressive overall survival and progression-free survival benefit with AMG 510 compared with other standard therapies in the arsenal, Price concludes.
