This multinational, randomized, phase III trial of the combination of capecitabine and irinotecan (XELIRI) evaluated safety with or without bevacizumab (Avastin) versus FOLFIRI with or without bevacizumab as second-line therapy for metastatic CRC.

Results showed that bevacizumab did not change safety profiles in either the XELIRI or FOLFIRI arms. Both were both well tolerated, says Muro, with incidence of toxicities slightly lower in the XELIRI arm at 53.2% compared with 73% in the FOLFIRI arm.

Dr. Muro Discusses Safety Data from Asian XELIRI Project in CRC

Kei Muro, MD, Aichi Cancer Center Hospital, discusses the results of the PaFF-K study in metastatic colorectal cancer (mCRC).

This phase II multicenter study investigated the efficacy and safety of panitumumab (Vectibix) with FOLFOX or FOLFIRI as a first-line chemotherapy for KRAS-wild type mCRC. Of 162 total patients, 140 were treated with FOLFOX plus panitumumab and 22 with FOLFIRI plus panitumumab.

The overall response rate was 51.2% (95% CI: 43.3-59.2) and the disease control rate was 82.1% (95%CI:75.3-87.7) with a median follow-up of 28.2 months. Median progression-free survival and overall survival was 9.2 months for the FOLFOX arm and 33.8 months for the FOLFIRI arm. Maximum tumor shrinkage was observed at 10 and 16 weeks, respectively, in 115 patients.

	Kei Muro, MD, of the Aichi Cancer Center Hospital in Nagoya, Japan, discusses updated findings from the KEYNOTE-012 study, which looked at pembrolizumab for the treatment of several types of cancer.

	Muro recently presented on the gastric cancer cohort of KEYNOTE-012, which included 39 patients whose tumors expressed PD-L1. The study included 19 Asian and 22 non-Asian patients.

	The objective response rate (ORR) was 22.2% and the toxicity profile was very manageable. Median progression-free survival (PFS) was 1.9 months and at 6 months, the PFS rate was 24%, and the overall survival (OS) rate was 69%.
